Transmitted Darkfield Illumination
This is one of the series on Illumination of fungae. Here the object is made visible by
the light
that is normally lost to create the image. What was dark in the brightfield image is now
bright.
The contrast is good, though sometimes too high, and resolution is enhanced. Often a
phase contrast
plate in the condenser can be used as a darkfield condenser stop. As an example, the
phase plate for
a 40X objective may be a darkfield stop for a 10X objective, the phase plate for a 20X
objective may
be a darkfield stop for a 4X objective. If you have a phase condenser a little
experimentation can
produce some very interesting effects.
