Phase Contrast Dispersion Staining
This is a collection of glass fiber that has a
refractive index of 1.550 in a high dispersion liquid with
a refractive index of 1.545.
Transmitted Negative Phase Contrast Illumination
Definition/Function:
With negative phase contrast illumination the interior of the particle shows the
wavelenghts for which the mounting medium has the higher refractive index and the halo
shows the wavelengths for which the particle has the higher refractive index. If the
refractive index of the particle and
the mounting medium match at some visible wavelength but the dispersion curve of the
mounting medium has a greater slope than that of the
particle then the partile and the halo become colored.
